Online measurement data of continuous aqueous two-phase flotation (atpf) for enzyme purification

TechnicalRemarks: - The .csv files contain experimental data obtained from ATPF experiments at laboratory scale. - The flotation tank used contains 700 ml of the aqueous two-phase system (ATPS). The phase forming components were polyethylene glycol 1000 and trisodium citrate dihydrate. - Phospholipase A2 was used as the model enzyme and the model suspension was mixed with the bottom phase. - The flotation tank was conceptually divided into three compartments, each compartment containing a gassing unit and an electrical conductivity probe. The electrical conductivity probes were inserted 1 cm below the phase interface. - An UV/Vis spectrometer with flow cell was implemented at the top phase outlet to obtain the top phase concentration in real-time. The bottom phase concentration was calculated via a mass balance equation.

  • The file names contain the process parameters in the following order: feed concentration_gas flow rate combination (Vd_1-Vd_2-Vd_3, see also below)_residence time_volume ratio (top to bottom phase)_experiment number.

  • R_top, R_bot: top and bottom phase recirculation (R = 1 for full recirculation, R = 0 for no recirculation)
  • c_bot_3, c_top_3: enzyme concentration obtained from UV/Vis measurements in % (w/w).
  • kappa_bot_1, kappa_bot_2, kappa_bot_3: Electrical conductivity measurements in all three compartments in mS/cm.
  • Qd_bot, Qd_top: Volume flows of top and bottom phase in ml/min.
  • Vd_1, Vd_2, Vd_3: Gas volume flows of the three gassing units in ml/min.
